Isolation of Legionella anisa using an amoebic coculture procedure.

Conventional diagnostic tests for legionellosis were negative for a 61-year-old immunocompromised man with pneumonia. However, coculture of a sputum sample with Acanthamoeba polyphaga amoebae led to the recovery of Legionella anisa. This procedure may be a sensitive and convenient diagnostic method, especially for non-Legionella pneumophila species infections that can be diagnosed only by culture.

Legionella anisa is a gammaproteobacterium from the class Legionellaceae, which is responsible for nosocomial pneumonia. We sequenced the genome from the L. anisa strain Linanisette, which was recovered from a clinical sample using an amoebal coculture procedure but not with standard culture methods.

Viable Legionella pneumophila bacteria were isolated by amoebal coculture from pluvial floods after intense rainfall and from water collected at sewage treatment plants. Several isolated L. pneumophila strains belonged to sequence types that have been previously identified in patients.
